1) Davros cannot be the Doctor's brother, as he is a Kaled, whereas the Doctor is a Time Lord.
2) It was established in 1963, and again in 2006, that The Doctor has a family. His mother was a human, his granddaughter (Susan) is human.
3) It was believed, although to my knowledge unconfirmed, that the Doctor and the Master are cousins / half-brothers. They certainly attended the Academy together.

Whether Susan was Human or Gallifreyan was never established. I've seen many theories relating to Susan relationship with The Doctor, and why she called him "Grandfather", and any of them or none of them could be right. The only way we'll find out is if we ever encounter the character again.

That line was indeed said by The Master in The Five Doctors when Doctor 1 asked The Master if he knew him. Now there is a reason why Doctor 1 might not have recognised him, and that's because in "The Keeper Of Traken", the Master stole the body of Tremas, so the Doctor would not have recognised the body before him as Gallifreyan, and rightfully so.
There is another possibility.
As the end of Season 11 of the original series, the original plan was a final confrontation between the Doctor and The Master, where it would be revealed that the Master was really The Doctor's dark side. Id to the Doctor's ego as it were.
At the end of the story, the Master would have died in a big explosion, saving others, including The Doctor. But why he had done this, would be left completely ambiguous.
Writer Robert Sloman was hired to write the story, which was initially titled, "The Final Game". However, it never got beyond the outline stage, as tragically Roger Delgado, who played The Master, was killed in a car accident in Turkey.
The loss of his great friend, and the departure of Katy Manning from the series the previous year, convinced Jon Pertwee that it was time to move on from the role, and so, in the story that Sloman wrote to replace it, Planet Of The Spiders, the Doctor 'died' instead of the Master. It is worth noting that for "Planet Of The Spiders", the voice of the Queen Spider is provided by Roger Delgado's widow, Kismet.
If that was the case, then what Doctor 1 did in The Five Doctors would have been no different from what he had done all along, and that was refuse to recognise his own dark side. The concept was later borrowed, to a degree, for the character of The Valeyard in Trial Of A Time Lord.
If the Doctor is 50% human (on his mother's side) then, even if whoever Susan's grandmother is was 100% human, that would still make the Doctor's offspring (i.e. Susan's mum/dad) one-quarter Time Lord.
Therefore Susan herself - even if her other parent is 100% human - must be at least one-eight Time Lord, rather than
totally
human. Which probably isn't enough to afford Susan certain Time Lord traits, like the ability to regenerate, though.
Will this forthcoming "son-of-the-Doctor" be meant to be Susan's dad? Or a different offspring of the Doctor?
If the former, then he shouldn't be played by a kid IMHO, but perhaps someone virtually the same age as David Tennant (or perhaps older!), so as to make a point of the fact that how old a Time Lord
looks
(in human judgement) bears no relation to his/her actual age.
Then again, it's possible that the Doctor had a relationship long after Susan had already grown up, so actually has a son who's younger than his grand-daughter! In which case, the forthcoming son being played by a kid would be acceptable.
PS: Who the f*ck is Charlie Clements?
EDIT:
Just Googled for Charlie Clements. Oh, the ginger lad out of EastEnders! (I've
never
been an EastEnders watcher. I'm only vaguely aware of it, and him, through friends that watch EE). He's rather fancyable. Drool...
Are you sure that's the case? I know it was mentioned in the US TV movie but I wouldn't be surprised if they didn't just pretend it was never said - it hasn't been mentioned since (to my knowledge).
Oh and am I missing something - is David Tennant leaving?! If not, what's the point of this thread?!?
maybe they want charlie clements to the the next doctor because in the christmas invasion episode the doctor complains about never being ginger.
Now that both Pauleen Fowler and Kathy Beale in Eastenders are dead it messes up the Dr Who continuity from the 30th anniversary special from 1993. Different doctors appear in Albert Square in different years and the 3rd Doctor (Jon Pertwee) ends up there in 2013 and asks Kathy and Pauleen what year it is.

There's been some speculation by The Sun, but there's been no definite announcements.

Well, it was a Children in Need special, so probably shouldn't be taken too seriously (otherwise you'd have to ask why the Tom Baker Doctor had grey hair, for instance).
Rationalise it away as the Rani's interference sending them into a parallel reality.
